Scientists from Azerbaijan and Italy offer a method of therapeutic treatment of cataracts in diseases such as Alzheimer’s, Parkinson’s

Recent studies have shown that amyloid protein diseases such as Alzheimer’s, Parkinson’s, etc. are very common among the world’s population.

These diseases belong to the class of age-related diseases. Many other diseases and their complications, such as cataracts, diabetes, etc., are associated with the aggregation of proteins and the formation of amyloid. It is expected that in the near future, amyloid diseases will become a serious medical and economic problem in the world.

Joint research conducted within the framework of a joint program under the cooperation agreement between the National Academy of Sciences of Azerbaijan (ANAS) and the National Research Council of Italy (CNR) has opened up new prospects in the fight against amyloid diseases.

On the Azerbaijani side, the research is led by the director of the Institute of Biophysics of ANAS, corresponding member Oktay Gasimov, and on the Italian side – a scientist of the Institute of Chemical Sciences and Technologies (Milan) Laura Ragona. For a long time, the Ph.D. student of the Institute of Biophysics Aida Mammadzade took an active part in the research work.

Silk fibroin (SF) is a non-pathological amyloidgenic protein prone, in solution, to the formation of amyloid-like aggregated species, displaying similarities in fibrillation kinetics with pathological amyloids, as widely reported in the literature.  Experts show here, on the basis of different biophysical approaches (turbidity, Congo Red assays, CD, DLS and fluorescence), that fusidic acid (FA), a well-known antibiotic, acts on SF as an anti-aggregating agent in a dose-dependent manner, being also able to revert SF aggregation. FA binds to SF inducing changes in the environment of SF aromatic residues. We further provide the proof of principle that FA, already approved as drug on humans and used in ophthalmic preparations, displays its anti-aggregation properties also on lens material derived from cataract surgery and is capable of reducing aggregation. Thus it is suggested that FA can be foreseen as a therapeutic treatment for cataract and other protein aggregation disorders.

Fusidic acid, a known antibiotic, interferes with silk fibroin self-aggregation and is also effective in disaggregating human cataract lens material. Fusidic acid shows a therapeutic potential in cataract prevention. Silk fibroin, an easily available amyloidogenic protein, could represent a useful and inexpensive platform to select aggregation-inhibitors of pathological amyloids.

The important results obtained within the framework of the project were reflected in an article published in the International Journal “Biophysical Chemistry” with a high impact factor.

Azerbaijani and Turkish scientists have achieved significant results in the fight against Covid-19 and cancer

Recently, extensive research has been conducted on anti-virus and anti-cancer peptides in the fight against viruses and cancer. In addition to the fact that bioactive peptides are not toxic to the human body, they also have very effective anti-viral and anti-cancer properties.

According to the Cooperation Agreement signed between the Azerbaijan National Academy of Sciences (ANAS) and the Turkish Scientific and Technological Research Council (TUBITAK) as a result of joint research conducted under the Joint Program, the cationic EQRPR peptide obtained on the basis of hydrolysis of rice bran protein could be used in the fight against cancer and Covid-19 infection, and the mechanisms of their action were studied.

The research is led by the director of the National Oncology Center, academician Jamil Aliyev, director of the Institute of Biophysics, corresponding member of ANAS Oktay Gasimov, professor of Baku State University Niftali Gojayev, and professor Sevim Akuyuz from Turkey.

Both theoretical (molecular docking, molecular mechanics) and experimental (Fourier Transformer Infrared Spectroscopy, Raman Spectroscopy, Circular Dichroism and Electron Paramagnetic Resonance Spectroscopy) methods are widely used in complex research. In addition, both point D-isomer changes of the EQRPR peptide and complete D-isomer peptides were synthesized and changes in their pharmacological properties were studied.

It should be noted that scientists of the two countries for the first time using the method of molecular docking showed complexes of peptides and their D-isomer analogs with ACE2, SARS-CoV-2 virus with S- and Mpro proteins, as well as possible clinical profiles, were determined by calculating the combination constants of the complexes. The EQRPR peptide studied has been shown to be multifunctional.

According to experts, the peptide can bind to a number of important integrins. It can be widely used in the fight against both SARS-CoV-2 virus infection and cancer.

One of the important results is that the multifunctional peptide can be modulated by carrying out D-isomer changes at different points in the binding property of different proteins. The secondary structure of the EQRPR peptide was calculated by various spectroscopic methods, and the spatial structure was calculated on a theoretical basis. The successful combination of experimental and theoretical results is commendable.

Preliminary studies in lung cancer cells have shown that the EQRPR peptide interacts with the cell membrane. Important results from the project were published in a high-impact international magazine “Journal of Molecular Graphics and Modeling” (Evaluation of anti-cancer and anti-covid-19 properties of cationic pentapeptide Glu-Gln-Arg-Pro-Arg, from rice bran protein and its d-isomer analogs through molecular docking).

In conclusion, it should be noted that research work of significant clinical and fundamental importance is underway.

At the Institute of Biophysics of ANAS the scientific  report of the young scientist was heard

On April 9, 2021, at the Institute of Biophysics of ANAS the next online scientific seminar  was held.

The seminar was opened by the general director of the institute, corresponding member of ANAS Oktay Gasimov, who gave information about the young scientist to the participants of the seminar.

At the seminar, the report of the junior research scientist of the laboratory Structure, dynamics and function of biomolecules, PhD. student Laman Suleymanova on the topic ”Structural-dynamic changes occurring in protein monolayers”  was heard.

In her report, she spoke in detail about the monolayer protein and its production, the principle of operation of the Langmuir-blodgett device, the effect of Cu2 + ions on monolayer fibroin, experiments conducted on the topic and the results obtained. According to the young scientist, all biological processes occurring in organisms occur either directly through the process of regulation, in which proteins participate, or by their regulation. The various functions performed by proteins are closely related to their molecular structure and dynamics. Therefore, one of the most important problems of biophysics is the study of the relationship between the structure, dynamics and function of proteins.

She noted that widespread neurodegenerative diseases all over the world, such as Alzheimer’s, Parkinson’s, Prion, etc. occurs as a result of amyloid formation and stressed the importance of discovering the mechanisms of amyloid release and exploring ways to prevent it. L.Suleymanova noted that during the joint work in the laboratory, the effect of Cu+2 ions on the fibroin protein was studied and the presence of cu+2 ions in the composition of amyloids was found.

At the end of seminar, the discussions around the topic were held and the questions were answered.

29 years pass since Khojaly Genocide

“The Khojaly genocide with its inconceivable
cruelty and inhuman punitive methods,
was completely targeted against the people
of Azerbaijan and represents a barbarian act
in the history of humankind. At the same time,
this genocide was a historical crime against humanity.”

Heydar Aliyev, national leader of Azerbaijan

29 years pass from the Khojaly Genocide, an integral part of the long-lasting aggression and ethnic cleansing policy of Armenia against Azerbaijan, resulted in mass and gross violation of international norms and principles, human rights and freedoms, engraved into the world history as a bloody tragedy. This massacre is another horrible evidence of the genocide policy of the Armenian nationalists and their supporters against the Azerbaijani people.

During the Khojaly genocide, 613 people were killed, 1,275 civilians were captured, and the fate of 150 of them is still unknown.

Throughout history, Armenian nationalists have committed bloody crimes, acts of terror and genocide against the Azerbaijani people.  However, the Azerbaijani people bravely passed these ordeals. As a result of the 44-day Patriotic War led by wise Azerbaijani President, Victorious Commander-in-Chief of the Armed Forces Ilham Aliyev, Karabakh was liberated from the occupation and the people’s 30-year longing for the native land was ended. Our victorious army has avenged the innocent victims of this massacre.

As the staff of İnstitute of Biophysics, we respectfully bow our heads before the memory of all our martyrs! We will always be grateful to them, they will always live in our hearts!

The report of the young scientist was heard

On February 5, the Laboratory of Integrative Biology of the Institute of Biophysics of ANAS organized an online seminar on the topic “Abstract of alternative proteomes obtained as a result of alternative splicing and frameshifting”.

At the online seminar, which was organized on the “Zoom” platform, the junior researcher of the Laboratory of Integrative biology, the winner of the double doctoral program organized jointly by ANAS and Montpellier University of France for the 2019-2022 academic years, Zarifa Osmanli spoke on the topic.

She noted that the main research area of the Laboratory of Structural Bioinformatics and Molecular Modeling (Institute of Cell Biology, CNRS) is the study of the structure and functions of proteins using bioinformatics programs based on their amino acid sequences. Emphasizing that there are two main regions of proteins, the globular and the non-globular, she noted that the non-globular, in turn, are divided into aggregation-prone motifs, intrinsically disordered regions and tandem repeating subgroups. She informed that the laboratory is currently creating, improving and integrating bioinformatics programs for a large-scale analysis of the listed areas and subgroups, and also pointed out that the main focus of her Ph.D. is the study of dark proteomes using existing bioinformatics programs and writing new programs.

According to the reporter, many mechanisms of transcription and post-transcription lead to the formation of new proteins and isoforms of these proteins. Among these mechanisms, a special place is occupied by the mechanisms of “Alternative splicing” and “frameshifting”. The structural and functional analysis of alternative proteomes obtained by these mechanisms is the main part of the study. Abstract alternative pre-mRNA proteomes are a widespread mechanism that absorb a variety of proteins in eukaryotic organisms. Comparative analysis of isoform and reference protein sequences is carried out taking into account the possibility of obtaining several isoform proteins formed from a single coding region. Abstract of alternative proteomes is used for in-depth study of genome coding regions, identification of potentially new proteins, and recognition of genome regions that are more affected by evolution. The young scientist also stressed that until recently, it was discovered that “frameshift”, which only leads to the appearance of mutant proteins or premature stop codon, was already a method used to create new proteins in the process of evolution.

After hearing the report,  discussions were held on the subject. The General Director of the Institute, corresponding member of ANAS Oktay Gasimov, deputy director for scientific affairs, Dr. Biology Karim Gasimov, head of the laboratory of Integrative biology, corresponding member of ANAS Ilham Shahmuradov, head of the laboratory Molecular Basis of Integrative Activity of the institute  of Physiology  Arif Mehdiyev and others took part in the discussions.

31 years pass since the January 20 Tragedy

Thirty-one years have passed since the events of January 20, 1990, which are went into history as the tragedy of “Black January”.

On the night of January 20, 1990, the former Soviet military troops attacked Baku and carried out the punitive measures with unprecedented cruelty. During the tragedy the courageous sons of the Azerbaijani people who stood up for the motherland’s dignity, honour and freedom became martyrs.

We bow before the souls of our martyrs who gave their lives in Bloody January of 1990 for the freedom of Azerbaijan, and we remember them with deep respect. May God have mercy on all our Martyrs. The memory of our Martyrs will always be a saint for us.

Scientists of the Institute of Biophysics of ANAS have developed a method for characterizing proteins in solid form

The article of the young scientist of the Institute of Biophysics, Ph.D. Aytaj Mamedova and the director of the institute, corresponding member of ANAS Oktay Gasimov has been accepted for publication in the prestigious journal “Biochemistry and Biophysics Reports” published by “Elsevier”.

In the article entitled “ANS fluorescence: Potential to discriminate hydrophobic sites of proteins in solid states”, it is possible to characterize hydrophobic clusters of proteins in the solid form for the first time using the fluorescence properties of the ANS molecule (1-anilino-8-naphthalenesulfonate). Silk fibroin from Bombyx mori cocoons was used as a paradigm protein. ANS incorporated into the films of silk fibroin exhibits fluorescence with two-lifetime components that can be assigned to the patches and/or cavities with distinct hydrophobicities. Decay associated spectra (DAS) of ANS fluorescence from both sites could be fit to the single log-normal component indicating their homogeneity. ANS binding sites in the protein film are specific and could be saturated by ANS titration. ANS located in the binding site that exhibits the long-lifetime fluorescence is not accessible to the water molecules and its DAS stays homogeneously broadened upon hydration of the protein film. In contrast, ANS from the sites demonstrating the short-lifetime fluorescence is accessible to water molecules. In the hydrated films, solvent-induced fluctuations produce an ensemble of binding sites with similar characters. Therefore, upon hydration, the short-lifetime DAS becomes significantly red-shifted and inhomogeneously broadened. The similar spectral features have previously been observed for ANS complexed with globular proteins in solution. The data reveal the origin of the short-lifetime fluorescence component of ANS bound to the globular proteins in aqueous solution. Findings from this study indicate that ANS is applicable to characterize dehydrated as well as hydrated protein aggregates, amyloids relevant to amyloid diseases, such as Alzheimer’s, Parkinson, and prion diseases.

Long live the Azerbaijani Army!

Nagorno-Karabakh region and seven surrounding districts of Azerbaijan have been under occupation for almost 30 years.  From the four known UN Security Council resolutions to various international organizations, the entire civilized world has repeatedly stated in official legal documents that Karabakh is the territory of Azerbaijan and occupied by Armenia.  Despite all this, the armed forces of the Armenian state, which did not fulfill its obligations under international agreements and did not comply with them, remained committed to the policy of aggression, and from September 27, 2020, again grossly violated the norms of international law, opened artillery fire on the densely populated areas of Azerbaijan.  In response to the Armenian provocation, the Azerbaijani Army, headed by the President, Supreme Commander-in-Chief Ilham Aliyev rose to fight for the liberation of our own lands.  As a result of counter-offensive operations, a number of our occupied villages and strategic heights were liberated from the enemy.

Each of us must unite in this difficult day and express support to our President, Supreme Commander-in-Chief Ilham Aliyev.  “It is the duty of each of us to do our best to bring the Great Victory Day closer!”, – these words of the first vice-president of Azerbaijan Mehriban Aliyeva are addressed to all our citizens.  In this sensitive and responsible period, every Azerbaijani, regardless of his identity and field of work, should try to be useful to his homeland.  Today, we must mobilize as one against the enemy’s information war, spread the truth about Karabakh in local and foreign media and social networks, fight against the disinformation spread by Armenia, and convey the true voice of Azerbaijan to the whole world.  We are on the right path! Ours is the cause of justice, we are fighting on our own lands.

The staff of the Institute of Biophysics of ANAS supports our brave army, which under the leadership of the Supreme Commander-in-Chief Ilham Aliyev conducts successful military operations to liberate the Azerbaijani lands occupied by the Armenian armed forces and wishes new victories to our army.

Victory is ours! Karabakh is Azerbaijan!

Appeal of the President of ANAS, academician Ramiz Mehdiyev

As is known, since September 27, 2020, the Azerbaijani Armed Forces have been fighting against the aggressor in the Armenian-occupied Nagorno-Karabakh and regions for almost 30 years, in response to a series of provocations by the Armenian army against civilians and military facilities in the frontline. From the first days of the operation, the Azerbaijani Army, demonstrating unparalleled courage and high professionalism, dealt crushing blows to the aggressor, liberated many settlements controlled by the Armenian military, as well as strategic heights. Thus, because of operations conducted from September 27 to October 9, Azerbaijan regained its sovereignty over the city of Jabrayil and its 6 villages, 7 villages of Fuzuli region, Sugovushan and Talish villages, Sur village of Khojavend and Hadrut settlement, as well as strategic heights in Murovdagh…

An employee of the Institute of Biophysics spoke at the Main Health Department

Head of the Laboratory of Ecological Biophysics of the Institute of Biophysics of ANAS, Dr. Biology, Professor Tokay Huseynov delivered a lecture at the Baku Main Health Department.

The report was devoted to research based on numerous studies on the impact of one of the environmental factors (selenium status) on the development and pathogenicity of HIV/AIDS, which is part of the Covid-19 genome, Ebola and other RNA viral infections.

Based on existing and scientifically proven research, Tokay Huseynov noted that the lack of selenium (Se) in human food, one of the chemical elements, can lead to the development of the most common viral diseases of our time. He said Hantavirus, Coxsackie, avian flu, Ebola and other dangerous viral diseases had symptoms similar to COVID-19.

The speaker also stressed that the increasing selenium deficiency in many countries, including Azerbaijan, is caused by a significant deterioration in the ecological situation, which is directly related to anthropogenic human activities in particular, the intensive development of energy, industry, communications, transport, which causes significant emissions of gas anhydrides into the atmosphere, soil acidity, as well as heavy elements, reduces the mobile forms of selenium absorbed by plants and entering the human body through them.

According to the professor, the use of selenium as a drug (mainly in the form of sodium Selenite) has led to a significant improvement in the condition of patients, and even complete recovery (Coxsackie virus – Keshan, China). He noted that selenium was one of the main drugs in South West Africa that prevented transmission of the disease from HIV to AIDS. In many countries around the world suffering from selenium deficiency, including developed countries and most of Asia, Australia, Africa, as well as in many CIS countries (Russia, Ukraine, Belarus, etc.) and the Baltic States, have adopted state programs to ensure the normalization of selenium status at the legislative level.

Speaking about the fact that Covid-19 is a vascular disease rather than a respiratory one, the speaker stressed that the tragic consequences of the virus were mainly due to blood clots, while sodium selenite had antithrombotic properties and prevented thromboxane formation.

According to the scientist, it was also found that thrombopenia, which is directly related to the development of stroke, occurs during infection with the COVID-19 virus: “All this suggests that the susceptibility to coronavirus and its complications depends on the degree of selenium intake into the body. Sodium Selenite itself can be effective in the treatment of this disease,” T. Huseynov said.

The enemy was given a decent response

While the whole world is fighting against one common enemy – COVID-19, once again Armenia does not hesitate to demonstrate its aggressive and terrorist intentions, and by its provocative actions grossly violates the fundamental norms and principles of international law continuing to encroach openly on sovereignty and territorial integrity of the Republic of Azerbaijan. Thus, according to the Ministry of Defense of the Republic of Azerbaijan, starting from noon on July 12, 2020, the Armenian armed forces violated the ceasefire in the direction of Tovuz region of the Azerbaijan-Armenian state border, and in order to exacerbate tensions, opened artillery fire on densely populated areas, taking attempt to attack. While preventing this provocation, the brave and courageous sons of Azerbaijan were heroically martyred.

However the blood of our martyrs was not go unrevenged. The enemy received a decent response and suffered significant losses of manpower and equipment.Currently, many countries around the world have shown support for our country, declaring that they will stand by Azerbaijan even in the most difficult times. The manifestation of friendship and solidarity by a number of countries at this sensitive moment has set another clear example of the correct foreign policy of the President of Azerbaijan.

In such a difficult time, all our citizens united and demonstrated their support for the Supreme Commander-in-Chief and the Armed Forces, as well as their resolute protest against the insidious and cunning aggression of the enemy.

The staff of the Institute of Biophysics, as well as all the Azerbaijani people, are always ready for each call and order of the Supreme Commander-in-Chief Ilham Aliyev.

We are strong together!

Oktay Gasimov, director of the Institute of Biophysics, corresponding member of ANAS

Studies of employees of the Institute of Biophysics against COVID-19  is included in the treatment protocol TABIB

Research carried out on the basis of existing data by employees of the Institute of Biophysics of ANAS- Prof. Tokay Huseynov and Ph.D. Ruhiyya Guliyeva on the impact of one of the environmental factors (selenium status) on the development and pathogenicity of HIV / AIDS, which is part of the COVID-19 genome, is included in the Protocol of the Administration of the Regional Medical Divisions (TABIB).

The results of studies aimed at finding methods of controlling virus COVID-19, have shown that a deficiency of selenium (Se) in human food, one of the chemical elements, can lead to the development of the most common viral diseases of our time.

Studies have shown that  the sensitivity to the virus COVID-19 and its implications seriously depend on the degree of selenium supply in the body, and the sodium Selenite itself is effective in the treatment of this disease.
